Characterization of bulge-specific DNA cleaver by matrix-assisted laser desorption inization time-of-flight mass spectrometry

摘要

The determination of reaction mechanism of DNA cleavage using matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) was discussed. The cleavage of HIV-27 was done by mixing 20 μM of HIV-27 and CoII complex in 20 μL H2O with 0.1% H2O2. The purification of HIV-27 and its cleavage products was done by ethanol precipitation, followed by denaturation. It was observed that the CoII complex was a bulge-specific probe to DNA structure, cleavaging at bulge site.
